CoatingProcedure

CellcultureinMebiol®flask:

  1. Openpackageinsterileenvironmentandadd10mLofculturemediadirectlytoMebiol®flask.
  2. Closeflaskandlayinrefrigerator(2-10°C)for3hourstoallowMebiol®toabsorbmedia.
  3. PeriodicallyshakeflaskgentlytohelpdissolvetheMebiol®intothemedia.Maketakeuptoadayforcompletedissolution(maintainatcoldtemperature).
  4. Afterdissolving,allowtositinrefrigeratoruntilbubblesareeliminated.
  5. Addcells/tissuesintoMebiol®atlowtemperature(2-10°C)andwarmto37°Ctoculturecells/tissuesina3Dhydrogelenvironment.
  6. Torecoverthecells/tissues,cooltheMebiol®backdownanddilutewith30-40mLofcoldsalineormedium.Thiswilltakeitoutofgelphase.SUSPendedcellscanbeeasilyrecoveredviacentrifugation.

Cellculturein24-wellplate:

  1. Followsteps1-4above.
  2. Beforeaddingcells/tissues,removedesiredamountofMebiol®andputintochilledcentrifugetube.PutremainingMebiol®intorefrigerator(stillinoriginalflask).
  3. Addapproximately105cell/mLtochilledcentrifugetubeandmixgentlyonice.
  4. Warmup24-wellplateandseparateoverlayingculturemediato37°C.
  5. Add200µLofcoldMebiol®suspensiontothecenterofeachwell.MakesureMebiol®doesnotcoverentirebottomofthewelltomakeiteasiertochangeoverlaidmediuminlaterstep.
  6. Overlay400µLofwarmedculturemediacontainingphenolredonMebiol®suspension.Cultureat37°C.
  7. Torecovercells,refrigerateat2-8°Canddilutewith400µLofcoldsalineormedium.ThiswilltakeMebiol®outofgelphase.Suspendedcellscanbeeasilyrecoveredviacentrifugation.

Donotre-sterilizetoavoiddeterioration.Addmediapromptlyafteropening,storeinrefrigerationandusewithin1month.
