HumanE-Cadherin(CDH1)isan882aminoacidprotein(1-23=signaldomain)thatbelongstoamemberofthecadherinsuperfamily.Theencodedproteinisacalciumdependentcell-celladhesionglycoproteincomprisedoffiveextracellularcadherinrepeats,atransmembraneregionandahighlyconservedcytoplasmictail.Mutationsinthisgenearecorrelatedwithgastric,breast,colorectal,thyroidandovariancancer.Lossoffunctionisthoughttocontributetoprogressionincancerbyincreasingproliferation,invasion,and/ormetastasis.Theectodomainofthisproteinmediatesbacterialadhesiontomammaliancellsandthecytoplasmicdomainisrequiredforinternalization.RecentpublicationfromDingSheng’sgroupindicatedthatcoatedrecombinanthumanE-cadherinprotein(extracellulardomain)alonebenefitshumanEScellCulture.
RecombinanthumanE-Cadheringene(155-710aaFragment)wasconstructedwithcodonoptimizationandexpressedinnon-fusionproteinforminE.coliasinclusionbodies.
