ElectrospunGelatinDiscsarefabricatedusingelectrospinningtechniquestocreateaconstructwithauniquestructuresuitableasabiomimeticthree-dimensional(3D)scaffold.Theproductcontainsoverlaidelectrospunfilamentscreatingaporousnetwork,whichpermitscellsandnutrientstoflowcompletelythroughtheporesandprovidesanincreasedsurfaceareaforcellattachment,growthandmigration.

ElectrospunGelatinDiscsarelightlycross-linkedforincreasedmechanicalstrengthanddurABIlityforshortandlong-termtissuecultureexperiments,yetisstillbiodegradableoverthelonger-term.Theaveragefilamentdiametertypicallyrangesfrom500nmto1.5microns.Theelectrospungelatindiscsareapproximately8mmindiameterand1-2mmthick.Theelectrospundiscsfitintoa48wellcultureplateorlarger.Eachpackagecontainsfivediscs.Thisproductissterilizedandready-to-use.

  1. PreparationandSeeding:

Note:Cellattachmenttothediscisgenerallythemostcriticalstepintissueculture.Temperature,pH,gasexchangeandcellconcentrationcanaffecttherateandefficiencyofattachment.Optimumseedingratedependsonthetypeofcellbeingcultured.

  1. AsepticallyremovetheElectrospunGelatinDiscsfromthepackaginginalaminarflowworkstation.
  2. Carefullyplacethediscsintothewellsofa48welltissuecultureplateorlargerusingasterileinstrument.Becarefulnottodamagetheproductasitisbeingtransferred.Itisrecommendedtousenon-treatedtissuecultureplasticware.Note:Tissue-coatedplasticwaremayneedtobecoatedwithagarosetopreventcellattachmenttotheplasticandpromoteattachmenttothedisc.
  3. SUSPendcellsatdesiredconcentration(approximately5,000cells/cm2)anddispensesufficientvolumeofcellsolutionontopofthediscplacedinthewell.

Note:Avoiddispensingthecellsolutiontoorapidlyasthismaycausedamagetothesponge.

  1. Transfertoa37ºCincubatorforabout1–2hourstoallowforinitialcellattachment.
  2. After1–2hour,removetheplatefromtheincubatorandcheckforcellattachment.Additionaltestingmayberequiredtooptimizethetimeittakesforthecellstoattachtothediscs.Checkthemorphologyofthecells.Celladherenceandspreadingwilldictatethetimeneededforattachment.
  3. Oncethecellshaveadequatelyattachedtothedisc,increasethefinalvolumeineachwelltofullycoverandprovideadequatemediumfortheculturesystem.
  1. ChangingtheMedia:
  1. Changethemedia24to36hoursaftertheinitialseeding.Thefrequencyofchangeswillbedeterminedbycelltype,cellattachmentefficiency,andpH.Morefrequentmediumchangesmayberequiredcomparedto2Dculturesystems.
  1. HarvestingofCells:

Note:Proteasedigestionisthestandardmethodofreleasingcellsfromthediscs.Thestrengthoftheattachmentofthecellstothediscswillvaryfromcelllinetocellline.Theenzymeconcentrationanddigestiontimewillvarydependingupontheactivityoftheenzymeandtheconfluenceofthecells.Collagenaseand/ortrypsinmaybethepreferredmethod.

  1. WashingthediscswithEDTA-PBSmayassisttheproteasedigestion.Addsufficientvolumetocoverthedisc.
  2. AspiratetheEDTA-PBSsolutionfromthewell.
  3. Addsufficientdissociationsolutiontothewelltofullysubmergethediscs.
  4. Transfertoa37ºCincubator.Checkforcelldetachmentperiodically.
  5. Oncethecellshavefullydetached,removethecellsanddispenseinacentrifugetube.
  6. Centrifugethecellsasrequired.
